U.S. Route 43 is a north–south United States highway that runs for 410 miles (649 km) from central Tennessee to Mobile, Alabama. The highway's southern terminus is in Mobile, Alabama, at U.S. Route 45. Its northern terminus is Columbia, Tennessee, at U.S. Route 31.

Between Mobile and Fayette and again from Killen to the Tennessee state line, U.S. 43 is paired with unsigned State Route 13. Between Fayette and Hamilton, U.S. 43 is co-routed with unsigned State Route 171. Between Hamilton and Florence, the unsigned partner for U.S. 43 is State Route 17. From its beginning in Mobile, Alabama, U.S. 43 passes through the western part of Alabama. It serves as the principal route connecting Mobile and Tuscaloosa, and from Tuscaloosa to Muscle Shoals and Florence. Prior to the completion of Interstate 65, motorists traveling from Mobile to Birmingham followed U.S. 43 to Thomasville; from there, they followed State Route 5 until its junction with U.S. Route 11 near West Blocton.
Except for Mobile, Tuscaloosa, and Muscle Shoals/Florence, U.S. 43 passes through rural areas and small towns.
Prior to the completion of Corridor X in western Alabama, U.S. 43 and U.S. 278 shared a concurrency with U.S. 78 between Guin and Hamilton.
